Service interlined with the 316 OSSINGTON night bus in order to provide more reliable running times. Southbound trips arriving at Springhurst continue through the Exhibition grounds by running south on Dufferin, east on Saskatchewan and east on Manitoba to the Exhibition streetcar loop, where the buses continue on as 316 OSSINGTON. In the opposite direction, 316 OSSINGTON buses arriving at Exhibition Loop change to 329 DUFFERIN and continue west on Manitoba, west on Saskatchewan and north on Dufferin, following the rest of the route to Lawrence station. When the Exhibition is closed for major events, the two routes are connected via Liberty, Frasier and King to Shaw.

September 4, 2005

Service is rerouted off of Lawrence to make way for the new 352 LAWRENCE WEST night bus. Buses on 329 DUFFERIN now follow the daytime route up Dufferin to Wilson and east on Wilson to Wilson station. Also on this date, accessible service is introduced using lift equipped Orion V buses, every day of the week.

A new Dufferin Street tunnel beneath the railway tracks at Queen Street is opened to traffic, allowing Dufferin and Dufferin Night buses to operate straight through the Queen/Dufferin intersection. Bus service removed from Peel, Gladstone and Queen.

July 12, 2013

Due to the Dufferin Street bridge over the Gardiner Expressway being declared unsafe and off-limits to motor vehicles, at least until a temporary bridge is opened ahead of a rebuild, service south of Dufferin Gate loop can not occur. However, as this route is interlined with 316 OSSINGTON NIGHT, the route is revised to find another way to Exhibition loop. After looping through Dufferin Gate loop, southbound buses operate north on Dufferin, east on Liberty, south on Strachan and then west via Manitoba Drive and the Exhibition loop access road where the buses start serving 316 OSSINGTON NIGHT. Returning buses proceed out of Exhibition loop and east on Manitoba Drive, north on Strachan, east on Liberty Street, south on Dufferin, then looping through Dufferin Gate loop again before heading north on Dufferin towards Wilson.

April 17, 2014

A temporary Dufferin Street bridge over the railway tracks at the Dufferin Gates is opened to car traffic, allowing service to resume south of Dufferin Loop. Night buses return to their previous routing to Exhibition Loop.

September 6, 2015

Service is extended at the north end to provide new service on Dufferin Street between Sheppard and Steeles Avenue. Buses follow the original route to Dufferin and Wilson and then operate east on Wilson, north on Transit Road, north on Allen, north on Dufferin, and west and north on Gerry Fitzgerald Drive to Steeles Avenue West. Buses return via south on Dufferin, south on Allen, south on Transit Road, west on Wilson and south on Dufferin to the Exhibition.

Ahead of the extension of the SPADINA SUBWAY to York University and into Vaughan, Downsview station is officially renamed Sheppard West. Destination signs have been adjusted in the days leading up to this change, although for 329 DUFFERIN, this only affects short turn runs.

March 27, 2022

The first two southbound trips of the 329 DUFFERIN NIGHT bus are diverted into Sheppard West station via Sheppard Avenue to pick up customers from the last YONGE-UNIVERSITY subway trains returning to Wilson Yar. These trips depart Steeles Avenue at 1:29 a.m. and 1:59 a.m. and arrive at Bay 2 of Sheppard West station at approximately 1:45 a.m. and 2:15 p.m.

Sunday, October 7, 2023

Service on this route is now operated out of Mount Dennis Garage. The following night, due to construction of the bridge at Dufferin Gates Loop, service is diverted via Liberty, East Liberty, Strachan, Manitoba Drive and Canada Boulevard to Princes Gates Loop.

Sunday, February 18, 2023

With the Dufferin Street bridge reopened, the regular service to Princes’ Gates is restored.
